2D cultures of specific cell types

iPS cell cultures can be differentiated into specific cell types. A number of robust differentiation protocols have been developed to generate highly purified cultures of progenitors as well as mature cell types from iPS cell lines.

Large scale experiments can be devised to generate monolayer (2D) cells from healthy and diseased iPS cells. These 2D cultures can contain specific cell types, such as a pure population of cardiomyocytes, or multiple cell types, such as cortical cultures which sustain the growth of neuronal and glial cell types.

The Stem Cell & Organoid Facility can generate 2D culture of cardiomyocytes, brain cells, microglia and hepatocytes (illustrated below).

Cortical Neurons

We have expertise in generating 2D neural progenitors (PAX6+/Nestin+), mature neurons (GABA+/VGLUT1+) and glial (GFAP+) populations.

Microglia cells

Microglia are the innate immune cells of the central nervous system. We can generate 2D pluripotent stem cell –derived microglia cultures (Iba1+) for various specifications.

The Facility has been working on optimising co-cultures of microglia with 2D cell types and organoids to form complex cultures containing multiple cell types of relevance.

Hepatocytes

The Facility optimises endoderm differentiation to each specific iPS cell line and works with customers to generate 2D iPSC-derived generate mature hepatocytes (HFN4a +/Albumin+) from their desired cell lines.

Hepatocytes, derived from our control iPS cell lines are functional secreting Albumin and showing CYP enzyme activity.
